What Is HPLC Purity?
HPLC (High-Performance Liquid Chromatography) is the industry-standard method for measuring peptide purity. In HPLC analysis, a peptide solution is introduced into an HPLC column packed with stationary phase material and pumped through under high pressure. Different compounds move through the column at different rates based on their polarity — separating into distinct peaks detected by a UV absorbance detector (typically at 214nm or 220nm for peptide bonds).
HPLC purity percentage = (area of primary peak) ÷ (total area of all peaks) × 100. Purity figures describe the proportion of the primary peak relative to total peak area; the remainder consists of deletion sequences, truncated fragments and process residues. Why purity matters for research: A peptide at 90% purity contains 10% unknown impurities. LC-MS Identity Confirmation
HPLC purity alone is insufficient for research-grade peptide verification. A compound can report high purity by HPLC and still be the wrong sequence — a scrambled sequence, deletion mutant, or entirely different compound will appear as a single clean HPLC peak if it elutes at a similar retention time.
LC-MS (Liquid Chromatography–Mass Spectrometry) confirms identity by measuring the molecular mass of the compound and comparing it to the theoretical mass calculated from the amino acid sequence. Agreement within instrument tolerance (typically ±0.5 Da) confirms the compound has the correct primary structure.
